The Drosophila brain membrane protein Slo, a member of the slow transporter (Slc12A) family, is regulated by phosphorylation/dephosphorylation through an unknown mechanism. Here, we report that in mature Drosophila brains, phosphorylated Slo is present in a tyrosine-phosphorylated state and reversibly localized in the plasma membrane. In vitro dephosphorylation studies showed that Slo is phosphorylated by a tyrosine kinase and dephosphorylated by a tyrosine phosphatase that is associated with the cytoplasmic region of Slo. Our studies indicate that Slo is not dephosphorylated by PTPX1 (ref. 2), a protein-tyrosine phosphatase recently reported to be associated with the cytoplasmic region of mammalian Shaker K+ channels.Q: Invertible element in $\mathbb{R}[x]$ Let $f(x) \in \mathbb{R}[x]$ be an irreducible polynomial of degree $3$ over $\mathbb{R}$ with root $x_0 \in \mathbb{C}$, such that $|x_0| eq 1$.
